Overview

GM.CD40L is a cell-based cancer vaccine composed of irradiated tumor cells that have been genetically modified to express both granulocyte-macrophage colony-stimulating factor (GM-CSF) and CD40 ligand (CD40L). This allogeneic tumor cell-based vaccine is designed to recruit and activate dendritic cells at the site of administration. The activated dendritic cells then migrate to regional lymph nodes where they stimulate T-cell activation, leading to systemic anti-tumor immune responses and tumor cell killing. The approach aims to enhance the immunogenicity of tumors by providing both antigen presentation (from the tumor cells) and potent immune stimulation via GM-CSF and CD40L expression. It has been investigated primarily for use in solid tumors such as non-small cell lung cancer (NSCLC), with clinical trials evaluating its safety, tolerability, and efficacy as an intradermal immunotherapy[5][6][8][9].
