Overview

GMA-161 is a humanized, aglycosyl monoclonal antibody that targets the human low-affinity Fc gamma receptor III (CD16), also known as FcγRIII. It was developed for the treatment of autoimmune disorders, specifically idiopathic thrombocytopenic purpura (ITP). The drug acts as an antagonist of CD16 and was designed to block the deleterious action of self-directed antibodies mediated by this receptor. Preclinical studies were conducted in transgenic mice expressing both isoforms of human CD16 (CD16A and CD16B), which supported its transition into phase I clinical trials. However, development was discontinued after early-phase clinical evaluation due to adverse reactions observed in preclinical models. The drug was originally developed by Genzyme Corporation and MacroGenics; later development efforts involved Sanofi following acquisition[5][6][7][8].
