Overview

GMA301 is a humanized monoclonal antibody that targets endothelin receptor A (ETA), a G protein-coupled receptor implicated in the pathogenesis of pulmonary arterial hypertension (PAH). By antagonizing ETA, GMA301 blocks the effects of endothelin-1, which is associated with increased vascular pressure and remodeling in PAH. Preclinical studies demonstrated that GMA301 lowers pulmonary arterial pressure and reduces right ventricular hypertrophy in animal models. The drug has shown an excellent safety profile and long half-life (up to 566 hours) in early clinical trials, supporting once-monthly intravenous administration. Developed by Gmax Biopharm, it is currently undergoing Phase 1 clinical trials for PAH in both children and adults. It was also previously under development for ovarian cancer and prostate cancer[1][2][3][4][5][6].
