GMcsf-FHAB-IL12 is a bispecific cytokine fusion protein developed by Sonnet BioTherapeutics using its proprietary Fully Human Albumin Binding (FHAB) platform. The molecule consists of Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF) and Interleukin-12 (IL-12) linked to an albumin-binding single-chain variable fragment (scFv). This design is intended to extend the serum half-life of the cytokines and enhance their delivery to tumor tissues by exploiting albumin-mediated transport pathways involving FcRn, GP60, and SPARC. By combining GM-CSF and IL-12, the drug aims to bridge innate and adaptive immunity, promoting the activation of dendritic cells, natural killer (NK) cells, and cytotoxic T lymphocytes within the tumor microenvironment. Preclinical studies in melanoma models have demonstrated significant tumor growth inhibition and a favorable safety profile compared to non-targeted cytokine therapies.
