GMI-1359 is a small molecule, glycomimetic compound developed as a dual inhibitor of E-selectin and CXCR4. Both E-selectin and CXCR4 are adhesion molecules implicated in tumor cell trafficking, metastatic spread, and the bone marrow microenvironment. By simultaneously inhibiting these targets, GMI-1359 aims to disrupt processes critical for cancer progression and metastasis. Preclinical studies suggested potential efficacy in cancers involving the bone marrow (such as acute myeloid leukemia and multiple myeloma), solid tumors that metastasize to bone (including breast cancer and prostate cancer), as well as rare pediatric tumors like osteosarcoma. The drug has received Orphan Drug designation for osteosarcoma[3][4][6].
