GMI-1757 is a novel, small molecule glycomimetic dual antagonist that targets both E-selectin and galectin-3, developed by GlycoMimetics. By inhibiting these two targets, the compound aims to address both the inflammatory and fibrotic components of various diseases. In preclinical oncology models, particularly pancreatic adenocarcinoma, GMI-1757 has demonstrated the ability to reduce intratumoral fibrosis and enhance immune cell infiltration, thereby improving the efficacy of checkpoint inhibitors like anti-PD-L1 therapy. Additionally, it has shown anti-thrombotic activity in models of deep vein thrombosis, demonstrating the ability to inhibit thrombus formation in inferior vena cava models. As of 2021, the compound was in preclinical development for the treatment of various cancers and fibrotic conditions.
