Overview

GMZ2 is a recombinant protein-based malaria vaccine candidate designed to prevent Plasmodium falciparum malaria. It consists of conserved domains from two asexual blood-stage antigens of P. falciparum—glutamate-rich protein (GLURP) and merozoite surface protein 3 (MSP3)—fused into a single recombinant fusion protein and adjuvanted with aluminium hydroxide (alum). The rationale for combining these two antigens is to mimic naturally acquired immunity against malaria by eliciting functional antibodies that target the blood stage of the parasite's lifecycle. Clinical trials have shown that GMZ2 is well tolerated, immunogenic, and induces both antibody and memory B-cell responses in both malaria-naive adults and individuals exposed to malaria. Phase 1 studies demonstrated safety and immunogenicity; phase 2 multicenter trials in African children showed modest efficacy in reducing the incidence of clinical malaria[4][6][7][9]. Further development may focus on improving its immunogenicity.
