Overview

GN-4474 is a live biotherapeutic product developed by Atterx Biotherapeutics for the prevention and treatment of infections caused by gram-negative bacteria. It utilizes a novel conjugation-mediated killing technology, where engineered bacteria carry a "killer plasmid" that is inactive due to repressors until transferred to pathogenic gram-negative bacteria via bacterial conjugation. Once inside the target pathogen, the plasmid becomes active and kills the host bacterium. This approach leverages gene transference as its mechanism of action and has demonstrated efficacy in preclinical models, including significant reduction of established infections and prevention of sepsis in animal studies. Notably, repeated exposure experiments have shown no evidence for resistance development among targeted pathogens[1][2][5][3]. The drug is being positioned as an alternative to traditional antibiotics for multi-drug resistant gram-negative bacterial infections.
