Overview

GN25 is a small molecule inhibitor that disrupts the protein-protein interaction between the tumor suppressor p53 and the transcription factor Snail (SNAI1). In many cancers, particularly those with K-Ras mutations or activated Hedgehog signaling, Snail is upregulated and binds to p53, effectively suppressing its pro-apoptotic and cell-cycle regulatory functions. GN25 blocks this binding, thereby re-activating the p53 pathway and inducing the expression of downstream targets such as p21 and Bax. Preclinical studies in pancreatic cancer models have demonstrated that GN25 can inhibit tumor growth and induce apoptosis, especially when used in combination with chemotherapy agents like oxaliplatin, regardless of the mutational status of p53 or K-Ras.
