GNC-039 is an octavalent, tetra-specific T cell engager antibody designed for cancer immunotherapy. It targets four tumor-associated antigens simultaneously—CD3, EGFRvIII (epidermal growth factor receptor variant III), PD-L1 (programmed death-ligand 1), and 4-1BB (tumor necrosis factor receptor superfamily member 9)—to redirect and enhance T cell cytotoxicity against cancer cells. By binding to CD3 on T cells and EGFRvIII on tumor cells, it facilitates targeted killing of EGFRvIII-expressing tumors. The engagement of PD-L1 allows the drug to overcome adaptive resistance mechanisms in tumors with high PD-L1 expression, while non-cytolytic engagement of 4-1BB provides a co-stimulatory signal that enhances T cell function over repeated dosing cycles.
