Drug intelligence / Profile preview
GNE684 (also known as GNE-684) is a potent, selective, and orally active small-molecule inhibitor of receptor-interacting serine/threonine-protein kinase 1 (RIPK1, also known as RIP1). Developed by Genentech, GNE684 acts as a type III allosteric inhibitor, blocking RIPK1 kinase activity to prevent necroptosis and apoptosis. In preclinical models, GNE684 has demonstrated therapeutic efficacy in reducing inflammation and tissue damage associated with colitis, ileitis, collagen antibody-induced arthritis, dermatitis, graft-versus-host disease (GVHD), and kidney ischemia-reperfusion injury (IRI).
