Overview

Gnetin C is a natural stilbenoid and a dimer of resveratrol, primarily extracted from the seeds of the *Gnetum gnemon* (Melinjo) plant. It is currently being investigated for its potent anticancer properties, particularly its ability to overcome therapeutic resistance in castration-resistant prostate cancer (CRPC). The compound acts by disrupting cellular energy metabolism; it downregulates Nicotinamide nucleotide adenylyltransferase 2 (NMNAT2), leading to the depletion of intracellular NAD+ levels. This depletion, combined with the suppression of key mitochondrial proteins and glycolytic enzymes, results in mitochondrial dysfunction and the inhibition of both glycolysis and oxidative phosphorylation. Gnetin C has demonstrated the ability to sensitize darolutamide-resistant cells to treatment, inducing apoptosis through the increased production of reactive oxygen species (ROS).
