Overview

GNOS-PV02 is a personalized therapeutic cancer vaccine (PTCV) developed by Geneos Therapeutics. It is a DNA plasmid-based vaccine encoding up to 40 patient-specific tumor neoantigens, designed to elicit strong immune responses against cancer cells bearing these unique mutated proteins. The vaccine is administered in combination with a plasmid-encoded interleukin-12 (IL-12) cytokine adjuvant and pembrolizumab, a PD-1 inhibitor. The mechanism involves the induction of tumor-infiltrating lymphocytes (TILs), particularly CD8+ T-effector memory cells with cytotoxic phenotypes, which target and kill tumor cells expressing the encoded neoantigens. Clinical trials have focused on advanced hepatocellular carcinoma (HCC), where the combination has demonstrated safety, immunogenicity, and promising efficacy as measured by objective response rate[1][2][3][4][5][6][8].
