GNP-LLO91-99 is a **nanovaccine comprising gold glyconanoparticles (GNPs) covalently coupled with the 91–99 amino acid peptide sequence from listeriolysin O**, a bacterial toxin. This nanovaccine is designed primarily as a cancer immunotherapy and adjuvant, promoting anti-tumor immune responses. In preclinical studies, GNP-LLO91-99 stimulates dendritic cell (DC) maturation and activation, enhances antigen presentation, and induces a Th1-type immune response. It triggers immunogenic tumor cell death and robust cytotoxic T-cell responses. GNP-LLO91-99 has demonstrated efficacy in reducing tumor burden and activating systemic anti-tumor immunity in murine models of melanoma and bladder cancer. It acts both as a **monotherapy** and in combination with immune checkpoint inhibitors (such as anti-CTLA-4 or anti-PD-1 antibodies), increasing CD4+ and CD8+ T cells and reducing immunosuppressive populations within the tumor microenvironment.
