GNP101 is an immunocytokine composed of the anti-HER2 monoclonal antibody trastuzumab fused to a mutein (modified version) of interferon-beta (IFN-β). Developed by GNP Bioscience, it is designed to treat HER2-positive malignancies, including those that have developed resistance to antibody-drug conjugates like trastuzumab deruxtecan (T-DXd). By targeting HER2, GNP101 localizes IFN-β to the tumor site, where it triggers direct anti-proliferative and apoptotic effects through the activation of the JAK-STAT signaling pathway, specifically inducing STAT1 phosphorylation. This dual-action approach combines the targeted binding of trastuzumab with the potent, non-cross-resistant cytotoxic activity of IFN-β, offering a potential therapeutic option for patients who have exhausted standard HER2-targeted therapies.
