GNTbm-TKI is a novel, orally administered **small molecule multi-tyrosine kinase inhibitor** with *potent immune-regulatory activity*, developed independently by Great Novel Therapeutics Biotech & Medicals Corporation (GNTbm). Its principal mechanism is the selective inhibition of multiple receptor tyrosine kinases: TYRO3, AXL, c-MER, BTK, ROS1, NTRK2, VEGFR2, and MET. GNTbm-TKI demonstrates strong immunomodulatory and tumor microenvironment (TME) remodeling capabilities. In preclinical models, it induces immune activation within tumors, suppresses tumor proliferation, invasion, and metastasis, inhibits angiogenesis, and is uniquely effective at transforming 'cold' tumors into 'hot' tumors when combined with HDAC inhibitors, immune checkpoint inhibitors (ICI), or anti-PD-1/VEGF bispecific antibodies. Its primary applications are: - Monotherapy for advanced neuroendocrine tumors - Combination therapy for a variety of advanced solid tumors, notably aiming to address unmet needs in tumors with immunologically cold TMEs GNTbm-TKI is currently in preclinical development, with plans to enter Phase I trials as an oral agent for cancer immunotherapy, expected around 2027[1][2][7].
