Overview

GNTI-148 is an allogeneic engineered regulatory T cell (EngTreg) therapy being developed by GentiBio for the treatment of Type 1 Diabetes (T1D). As an "off-the-shelf" cell therapy, it is designed to provide a scalable alternative to autologous treatments by using donor-derived cells. The therapy involves engineering regulatory T cells to express a T-cell receptor (TCR) specific to pancreatic islet antigens, allowing the cells to home to the pancreas. Additionally, the cells are engineered with a proprietary signaling complex, such as a chimeric interleukin-2 (IL-2) receptor, and the master transcription factor FOXP3 to ensure the stability, potency, and persistence of the Treg phenotype in the inflammatory environment of the diabetic pancreas. The primary therapeutic goal is to suppress the autoimmune destruction of insulin-producing beta cells and restore long-term immune tolerance.
