Overview

GNUV205 is a bifunctional recombinant fusion protein developed by Genuv Inc. for the treatment of solid tumors. It consists of an anti-programmed cell death protein 1 (PD-1) antibody fused to an engineered interleukin-2 (IL-2) variant using the proprietary NuvoFc™ heterodimeric antibody platform. The IL-2 component is designed with a "no-alpha but attenuated beta-gamma" signaling profile, which selectively activates effector T cells (Teff) and natural killer (NK) cells while avoiding the stimulation of regulatory T cells (Treg) that express the high-affinity IL-2 receptor alpha subunit (CD25). This design aims to enhance anti-tumor immunity while minimizing systemic toxicities like vascular leak syndrome. Additionally, the anti-PD-1 domain exhibits increased binding affinity in the acidic (low-pH) tumor microenvironment and is cross-reactive between human and murine targets.
