GNX201-ADC is a next-generation antibody-drug conjugate (ADC) developed by GlycoNex in collaboration with PrecisemAb Biotech. It utilizes a "Pro-Antibody" or "Antibody Lock" platform where the antibody's variable region is fused to a masking peptide via a protease-cleavable linker. This design ensures the drug remains inactive in systemic circulation, minimizing off-target toxicity. Upon reaching the tumor microenvironment, overexpressed proteases cleave the linker, "turning on" the antibody to selectively bind tumor-associated glycan antigens (TACA). Once activated and bound, the ADC delivers a potent cytotoxic payload directly to the cancer cells. It is currently in preclinical development for the treatment of various solid tumors.
