GO-2317 is a first-in-class small molecule prodrug developed by General Oncology. It is a new chemical entity (NCE) designed to convert into hydroxocobalamin (Vitamin B12b) upon dilution. GO-2317 is primarily utilized as a key component of the GO-4 combination therapy, which also includes melphalan, BCNU (carmustine), and ascorbic acid (Vitamin C). This combination is intended to treat patients with inherited mutations in BRCA1, BRCA2, or PALB2 by inducing DNA damage, suppressing DNA repair pathways, inhibiting ATP/glycolysis, and activating PARP. It is currently being evaluated in Phase 1 clinical trials for metastatic pancreatic cancer, metastatic breast cancer, and other solid metastatic tumors.
