GO-M100B is a first-in-class antibody-drug conjugate (ADC) developed by GO Therapeutics that targets the tumor-specific Tn-glycoform of Mucin 1 (Tn-MUC1). This clean target approach utilizes a monoclonal antibody that selectively recognizes aberrantly truncated O-glycans (Tn-antigen) on the MUC1 protein backbone, which are present in malignant epithelial cells but absent from normal tissues. The ADC is engineered as a site-specific conjugate with a drug-to-antibody ratio (DAR) of 2, utilizing an mc-vc-PAB linker and a monomethyl auristatin E (MMAE) payload. Preclinical studies have demonstrated potent cytotoxicity against Tn-positive cell lines and significant tumor regression in xenograft models of breast, lung, ovarian, pancreatic, and gastrointestinal cancers, while maintaining a favorable safety profile in non-human primates.
