Go-PRAME TCR T cells (BPX-801) is an autologous T-cell receptor (TCR) engineered T-cell therapy developed by Bellicum Pharmaceuticals. The therapy is designed to target the Preferentially Expressed Antigen in Melanoma (PRAME), a cancer-testis antigen that is highly expressed in various solid tumors but has limited expression in normal tissues. Go-PRAME incorporates Bellicum's proprietary GoTCR technology, which features an inducible MyD88/CD40 (iMC) co-stimulatory switch. This switch is designed to be activated by the administration of rimiducid (AP1903), a small-molecule dimerizing agent. Upon activation, the iMC domain provides potent co-stimulatory signals that enhance the expansion, persistence, and anti-tumor activity of the TCR-T cells within the immunosuppressive tumor microenvironment. Development of the program was discontinued following Bellicum's decision to halt its internal research and clinical operations in 2023.
