GoCAR-NK is an allogeneic engineered **CAR-NK cell therapy** platform originally developed by Bellicum Pharmaceuticals and later associated with intellectual property acquired by MD Anderson Cancer Center. The platform uses donor-derived natural killer cells engineered to combine antigen-directed chimeric antigen receptor killing with intrinsic NK-cell cytotoxicity. A distinguishing feature is an inducible MyD88/CD40 activation switch that can be triggered by rimiducid to enhance NK-cell activation, proliferation, persistence, and antitumor activity, together with a rapamycin-inducible caspase-9 safety switch intended to allow controlled elimination of the cells if needed. Reported GoCAR-NK programs have included constructs directed against BCMA for multiple myeloma, CD123 for acute myeloid leukemia, and HER2 for solid tumors, with autocrine IL-15 support designed to improve in vivo function and persistence.
