goCAR-T is a proprietary cell therapy platform developed by Bellicum Pharmaceuticals designed to treat solid tumors by incorporating an inducible costimulatory (iMC) switch. The technology utilizes T cells engineered with a chimeric protein consisting of MyD88 and CD40 signaling domains, which are activated only upon the administration of rimiducid, a small-molecule dimerizing agent. This "GO" switch allows for controlled T-cell activation, proliferation, and persistence within the immunosuppressive tumor microenvironment, potentially enhancing efficacy while mitigating the risks of constitutive CAR-T signaling. The platform has been applied to several clinical-stage assets, including BPX-601 (targeting PSCA) and BPX-603 (targeting HER2), and is often paired with a safety "STOP" switch (such as CaspaCIDe) to allow for the rapid elimination of engineered cells if toxicity occurs.
