Overview

Golcadomide is an orally available small molecule drug classified as a cereblon E3 ubiquitin ligase modulator (CELMoD). It is designed to enhance targeted protein degradation by binding with high specificity and affinity to cereblon (CRBN), the substrate recognition component of the CRL4-CRBN E3 ubiquitin ligase complex. This interaction leads to the ubiquitination and subsequent proteasome-mediated degradation of key proteins involved in cancer cell growth and survival, particularly in B-cell malignancies. Golcadomide exerts both direct cytotoxic effects on malignant B cells (inducing apoptosis) and immune-stimulatory activity in T cells. It represents a next-generation advancement over classic immunomodulatory drugs (IMiDs), offering improved efficacy even in IMiD-resistant settings. The drug is being developed primarily for various types of non-Hodgkin lymphomas, including diffuse large B-cell lymphoma, follicular lymphoma, primary mediastinal B-cell lymphoma, high-grade B-cell lymphoma, and transformed B-cell non-Hodgkin lymphoma.
