Overview

Golcadomide + poseltinib + rituximab is an investigational combination therapy being evaluated for the treatment of relapsed or refractory diffuse large B-cell lymphoma (R/R DLBCL). This regimen, studied in the Go-Pro-DLBCL trial sponsored by Seoul National University Hospital, combines three distinct therapeutic modalities to target B-cell malignancies. Golcadomide (CC-99282) is a potent, oral cereblon E3 ligase modulator (CELMoD) that induces the degradation of the transcription factors Ikaros (IKZF1) and Aiolos (IKZF3), which are essential for B-cell survival and proliferation. Poseltinib (HM71224) is a selective, irreversible inhibitor of Bruton's tyrosine kinase (BTK), a critical mediator of B-cell receptor signaling. Rituximab is a chimeric monoclonal antibody directed against the CD20 antigen on the surface of B-lymphocytes, promoting cell lysis through antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). The combination is designed to overcome resistance and enhance efficacy by simultaneously inhibiting multiple survival pathways in aggressive B-cell lymphomas.
