Gold nanoparticle + doxorubicin + Fab fragment is an experimental nanomedicine construct described in a 2023 academic study for the targeted treatment of prostate cancer. The system utilizes gold nanoparticles (AuNPs) as a core, which are coated with poly(propylene imine) (PPI) dendrons to enhance drug loading and facilitate lysosomal escape via the 'proton sponge' effect. The chemotherapeutic agent doxorubicin is conjugated to the dendrons through an acid-sensitive hydrazone bond, ensuring that the drug is primarily released in the acidic environment of the lysosome (pH 4–5). To achieve tumor-specific targeting, the nanoparticles are functionalized with a Fab fragment of an antibody that binds to the EphA2 receptor, which is overexpressed on the surface of prostate cancer cells. Preclinical evaluations in PC3 prostate cancer cells have demonstrated high cytotoxic potency, with an IC50 of 0.9 nM after 48 hours of incubation.
