Golidocitinib + corticosteroid is an investigational combination therapy being studied for the treatment of rapidly progressive interstitial lung disease (RP-ILD), particularly in patients with anti-MDA5 antibody-positive dermatomyositis. The regimen consists of golidocitinib (DZD4205), a potent and highly selective Janus kinase 1 (JAK1) inhibitor developed by Dizal Pharma, administered alongside standard-of-care corticosteroids (such as methylprednisolone or prednisone). Golidocitinib works by inhibiting the JAK-STAT signaling pathway, thereby suppressing the production of pro-inflammatory cytokines like interferon-gamma, which are central to the pathogenesis of RP-ILD. Corticosteroids provide complementary broad-spectrum anti-inflammatory and immunosuppressive effects. This combination is currently being evaluated in Phase 2 clinical trials to determine its efficacy in improving lung function and reducing mortality in this life-threatening condition.
