Overview

Golidocitinib (AZD4205) is a potent and highly selective Janus kinase 1 (JAK1) inhibitor. This combination therapy pairs golidocitinib with a programmed cell death protein 1 (PD-1) inhibitor for the maintenance treatment of patients with extensive-stage small cell lung cancer (ES-SCLC) who have achieved disease control following induction chemo-immunotherapy. JAK1 inhibition is thought to modulate the tumor microenvironment by reducing the signaling of pro-inflammatory and immunosuppressive cytokines, potentially overcoming resistance to checkpoint inhibitors and enhancing the anti-tumor activity of PD-1 blockade. The regimen is currently being evaluated in a Phase 2 clinical trial sponsored by the Cancer Institute and Hospital, Chinese Academy of Medical Sciences, with golidocitinib being developed by Dizal Pharma.
