Golnerminogene pradenovec is a recombinant oncolytic gene therapy agent consisting of a genetically modified adenovirus serotype 5 vector encoding the human cytokine tumor necrosis factor alpha (TNF-alpha). The drug is designed to deliver the TNF-alpha gene directly into tumor cells, leading to local production of TNF-alpha and subsequent stimulation of anti-tumor immune responses and induction of apoptosis in cancer cells. Its mechanism involves both direct cytotoxic effects via apoptosis pathways and immunomodulatory actions through enhanced cytokine signaling. Golnerminogene pradenovec was primarily developed for use in various solid tumors, including pancreatic cancer, head and neck cancer, melanoma, colorectal cancer, esophageal cancer, prostate cancer, and sarcoma[2][3][8]. The original developer was GenVec; later development involved Asahi Kasei and the National Cancer Institute (USA)[3][4].
