Gomisin M2 is a lignan isolated from the plant Schisandra chinensis (Turcz.) Baill., and also Schisandra viridis A. C. Smith[1][3][5]. It has demonstrated multiple pharmacological activities, primarily as an **anti-cancer** agent with notable effects against breast cancer stem cells, as well as **anti-allergic** and **anti-inflammatory** properties[1][2][3][5]. Mechanistically, gomisin M2 acts by inhibiting proliferation of triple-negative breast cancer cell lines and cancer stem cells, induces apoptosis through mitochondrial pathways and caspase activation, and downregulates Wnt/β-catenin signaling[1]. In allergic inflammation models, gomisin M2 suppresses mast cell degranulation and reduces the release of inflammatory mediators by inhibiting intracellular calcium influx and blocking FcεRI-mediated pathways, specifically the Lyn and Fyn kinases[2][3]. It also demonstrates efficacy in suppressing keratinocyte activation and pro-inflammatory cytokines in psoriasis-like models, likely involving inhibition of STAT1 and NF-κB pathways[4][5].
