Overview

Gonyautoxin is a marine toxin that belongs to the saxitoxin family. It exists in several forms (numbered as GTX1, GTX2, GTX3, GTX5, etc.) and has been investigated for medical applications, particularly for pain management. Gonyautoxin is a phycotoxin that works by reversibly blocking voltage-gated sodium channels at the axonal level, which impedes nerve impulse propagation[5]. This mechanism makes it potentially useful for pain management applications. The molecular structure is based on a 2,6-diamino-4-methyl-pyrollo[1,2-c]-purin-10-ol skeleton, with different gonyautoxin variants (GTX1, GTX2, GTX3, etc.) differing only by their substituents or stereoisomerism[3]. Gonyautoxin has been investigated in clinical trials for treating acute back pain and has shown promise in treating chronic tension-type headache[5]. In one study, patients reported significant pain relief within minutes after local infiltration of gonyautoxin, with no side effects detected during the follow-up period[5]. The compound is naturally produced by marine dinoflagellates like Gonyaulax and Protogonyaulax species and can be isolated from shellfish[9]. While it's naturally available, synthesis procedures for some gonyautoxin variants have been developed[3].
