GOP is an experimental triple-hormone combination therapy consisting of Glucagon-like peptide-1 (GLP-1), Oxyntomodulin (OXM), and Peptide YY (PYY). Developed primarily by researchers at Imperial College London, the therapy is designed to mimic the hormonal profile observed after bariatric surgery (specifically Roux-en-Y gastric bypass), which is known to lead to significant weight loss and rapid improvement in glucose homeostasis. The combination works through a multi-targeted approach: GLP-1 and OXM act as agonists at the GLP-1 receptor to enhance insulin secretion and suppress appetite; OXM additionally acts on the glucagon receptor to increase energy expenditure; and PYY acts primarily on the Y2 receptor to signal satiety. Clinical studies have demonstrated that subcutaneous infusion of GOP can significantly reduce food intake and body weight in patients with obesity and type 2 diabetes, leading to its description as a potential 'bariatric surgery in a bottle.'
