Overview

This combination regimen consists of goserelin, a gonadotropin-releasing hormone (GnRH) agonist, and the HER2-targeted monoclonal antibodies pertuzumab and trastuzumab. It is being evaluated by Peking Union Medical College in a Phase 2 umbrella trial (NCT05933408) for the treatment of salivary gland carcinoma, specifically for patients whose tumors are both androgen receptor (AR)-positive and human epidermal growth factor receptor 2 (HER2)-positive. Goserelin induces medical castration to suppress androgen-driven tumor growth, while trastuzumab and pertuzumab provide dual blockade of the HER2 receptor to inhibit dimerization and downstream oncogenic signaling. In the clinical trial context, this triplet is often administered alongside an AR antagonist to achieve comprehensive blockade of the AR and HER2 pathways.
