Overview

Goserelin + raloxifene is a combination of two pharmaceutical agents used experimentally for breast cancer risk reduction in premenopausal women at high risk. Goserelin is a synthetic analog of luteinizing hormone-releasing hormone (LHRH) that acts as a gonadotropin-releasing hormone (GnRH) agonist. It suppresses ovarian function and reduces estrogen production to postmenopausal levels. Raloxifene is a selective estrogen receptor modulator (SERM) that acts as an estrogen antagonist in breast tissue and an agonist in bone tissue. The rationale for combining these drugs is to achieve ovarian suppression with goserelin while using raloxifene to mitigate bone loss and provide additional anti-estrogenic effects on breast tissue[1][2]. This regimen has been studied for primary prevention of breast cancer in high-risk premenopausal women but is not standard therapy.
