Gossypol acetate is a small molecule polyphenolic compound derived from the cottonseed plant, currently being investigated for its potential to overcome chemotherapy resistance in oncology. It functions as a pan-histone deacetylase (HDAC) inhibitor and has been shown to specifically induce the degradation of the Leucine-rich pentatricopeptide repeat-containing (LRPPRC) protein. LRPPRC is a key mediator of resistance to chemotherapy in cancers harboring TP53 mutations. By targeting these pathways, gossypol acetate promotes apoptosis and autophagy in malignant cells. It is currently being evaluated in a Phase 2 clinical trial led by Guiying Wang as a second-line oral therapy in combination with bevacizumab and the FOLFIRI regimen, specifically targeting female patients with TP53-mutated and LRPPRC-positive metastatic colorectal cancer who have failed prior oxaliplatin-based treatments.
