gossypol acetate + bevacizumab + FOLFIRI is an investigational combination therapy being evaluated for the treatment of metastatic colorectal cancer. The regimen consists of gossypol acetate (an oral small molecule), bevacizumab (an intravenous monoclonal antibody targeting VEGF), and the FOLFIRI chemotherapy regimen (comprising irinotecan, 5-fluorouracil, and leucovorin). This specific combination is being studied as a second-line therapy for female patients whose tumors exhibit TP53 mutations and LRPPRC positivity. The scientific rationale is based on findings that LRPPRC (Leucine-rich pentatricopeptide repeat-containing protein) mediates chemotherapy resistance in TP53-mutated cells, and gossypol acetate can specifically degrade LRPPRC to reverse this resistance. Bevacizumab provides anti-angiogenic effects by inhibiting vascular endothelial growth factor (VEGF), while FOLFIRI provides standard cytotoxic chemotherapy through topoisomerase I and thymidylate synthase inhibition.
