Gotistobart is a next-generation, pH-sensitive anti-CTLA-4 monoclonal antibody. It is designed to selectively deplete tumor-infiltrating regulatory T cells by preserving CTLA-4 recycling in peripheral tissues, aiming to enhance anti-tumor immunity while reducing immune-related adverse events. This mechanism differentiates it from other CTLA-4 inhibitors by potentially offering an improved therapeutic index. Gotistobart is being co-developed by BioNTech and OncoC4 for various oncology indications, with a primary focus on metastatic non-small cell lung cancer.
