**GOVX-B21** is a preventive and therapeutic HIV vaccine candidate developed by GeoVax Labs using a DNA/MVA prime-boost platform. It expresses HIV clade B Gag, Pol, and Env proteins to form virus-like particles (VLPs) that mimic the virus structure, inducing robust T-cell and antibody responses. Distinct from GOVX-B11, GOVX-B21 co-expresses the genetic adjuvant **granulocyte-macrophage colony-stimulating factor (GM-CSF)** in the DNA prime to enhance immunogenicity, particularly for therapeutic use in HIV-infected patients on antiretroviral therapy. Evaluated in Phase 1 trials like HVTN 094 for prevention and planned follow-on studies for therapy, it showed good safety but no significant preventive benefit over the unadjuvanted version, leading to a shift toward advancing GOVX-B11.
