The gp100 DNA-HVJ-liposome vaccine is a preclinical cancer vaccine candidate designed to treat melanoma. It consists of a plasmid DNA encoding the human melanoma-associated antigen gp100 (glycoprotein 100) encapsulated within a fusogenic delivery system known as HVJ-AVE liposomes. This delivery vehicle combines the hemagglutinating virus of Japan (HVJ; Sendai virus) with artificial viral envelope (AVE) liposomes to facilitate efficient entry of the DNA into host cells via membrane fusion. Upon administration, the vaccine stimulates the production of gp100-specific antibodies and activates cytotoxic T lymphocytes (CTLs), which target and destroy melanoma cells expressing the gp100 antigen. Research published in 1999 demonstrated that intramuscular injection of this vaccine could induce protective immunity and significantly delay tumor progression in mouse melanoma models.
