Overview

The gp100-fowlpox vaccine is a recombinant viral cancer vaccine developed by the National Cancer Institute (NCI) Surgery Branch. It consists of a recombinant fowlpox virus (derived from the POXVAC-TC strain) engineered to express the gp100 (PMEL17) melanoma-associated antigen, a protein frequently overexpressed in melanoma cells. The vaccine is designed to stimulate and expand gp100-specific T-cell populations in vivo. In clinical research, it is primarily used as part of a multi-modal immunotherapy regimen for HLA-A*0201-positive patients with metastatic melanoma, typically following lymphodepleting conditioning and the infusion of adoptively transferred gp100-reactive or TCR-engineered lymphocytes, often in conjunction with high-dose interleukin-2 (IL-2).
