Overview

**gp100 peptides** are synthetic peptides derived from the tumor-associated antigen glycoprotein 100 (gp100), a transmembrane protein highly expressed in melanoma cells. These peptides are primarily employed as cancer vaccines, designed to stimulate cytotoxic T lymphocytes (CTLs) against melanoma by targeting peptide sequences presented by HLA Class I molecules (such as HLA-A*0201 or HLA-A*2402). Multiple formulations of gp100 peptides have been studied, including epitope-specific vaccines and liposome-encapsulated peptides[1][2][3][6]. Their mechanism of action involves the induction of antigen-specific immune responses, particularly activating CD8^+ T cells capable of lysing melanoma cells expressing gp100[2][3][7]. The strategy is commonly combined with immunoadjuvants or as a component of multi-modal melanoma immunotherapy. Clinical trials have demonstrated immunogenicity and safety, though objective anti-tumor responses are limited, suggesting these vaccines may be useful as part of combined immunotherapeutic regimens[2][3][6].
