gp100-specific CD8+ T cell clones are populations of cytotoxic T lymphocytes (CTLs) that have been selected and expanded ex vivo for their ability to recognize the melanoma-associated antigen gp100 (also known as Pmel17), a melanocyte differentiation protein highly expressed in many melanoma tumors. These cell clones are typically derived from the patient (autologous) or from donors and undergo adoptive cell transfer (ACT) as an investigational immunotherapy for metastatic melanoma. Their mechanism is direct antigen-specific cytolytic activity against gp100-expressing tumor cells. Studies have demonstrated that such clones can traffic to tumor sites, persist in patients, and mediate the regression of antigen-positive metastases with an acceptable safety profile, particularly when combined with low-dose IL-2[1][2][5].
