Overview

gp100-specific T cells are a form of adoptive T-cell therapy (ACT) designed to target the gp100 antigen, a membrane-associated glycoprotein (also known as PMEL) that is highly expressed in melanocytes and melanoma cells. These T cells are typically either isolated from tumor-infiltrating lymphocytes (TILs) or genetically engineered to express a specific T-cell receptor (TCR) that recognizes gp100 peptides presented by MHC molecules (most commonly HLA-A*0201). While primarily developed for the treatment of metastatic melanoma, research has explored their application in other gp100-expressing malignancies, including glioblastoma. The therapeutic approach relies on the ability of the transferred cells to migrate to tumor sites, infiltrate the tumor microenvironment, and execute effector functions against cells expressing the target antigen.
