gp100-targeted T cells are an adoptive cell therapy modality, typically involving T cells engineered to express a T-cell receptor (TCR) or chimeric antigen receptor (CAR) specific for the gp100 (PMEL) protein. gp100 is a melanocyte differentiation antigen that is highly expressed in most melanomas. These therapies are designed to recognize gp100-derived peptides presented by MHC molecules (in the case of TCR-T) or the surface protein itself (in the case of CAR-T), triggering T-cell activation and the subsequent destruction of melanoma cells. Historically, gp100-targeted TCR-T cells were among the first engineered T-cell therapies tested in clinical trials by the National Cancer Institute, demonstrating the potential for objective tumor regression in patients with metastatic melanoma, though often accompanied by on-target toxicities in healthy tissues containing melanocytes.
