gp100 TCR + B7H3 CAR T cells is an experimental dual-receptor adoptive immunotherapy being investigated for the treatment of pediatric high-grade gliomas (HGGs). This cell therapy involves genetically engineering T cells to co-express a T cell receptor (TCR) specific for the gp100 tumor antigen and a chimeric antigen receptor (CAR) targeting the B7H3 (CD276) antigen. The approach, termed TCR'CAR, aims to enhance T cell persistence, expansion, and cytotoxicity by combining native TCR signaling with CAR-mediated activation. Preclinical studies conducted by researchers at Children’s National Hospital have demonstrated that these dual-specific T cells exhibit superior sequential killing capacity and reduced exhaustion compared to single-receptor CAR T cells in HGG models.
