Overview

The **gp100 vaccine** is a therapeutic cancer vaccine designed to induce an immune response against tumors expressing the *gp100 antigen*, a melanocyte differentiation antigen commonly found in melanoma cells. Various formulations exist, including synthetic gp100 peptides, recombinant chaperone complex vaccines combining heat shock protein Hsp110 with gp100, DNA vaccines encoding gp100 (often with TRP-2), and liposomal formulations. The core mechanism is the stimulation of cytotoxic CD8+ T cell responses against melanoma by presenting gp100-derived peptides to the immune system, conferring tumor-specific immunogenicity. The vaccine is typically investigated in advanced or metastatic melanoma, both as monotherapy and in combination with other immunotherapies (such as interleukin-2, PD-1 inhibitors, or other checkpoint inhibitors).
