Overview

**gp100280-288 peptide** is an HLA-A2-restricted peptide vaccine (sequence YLEPGPVTA) derived from the melanocyte lineage-specific antigen gp100—also known as Melanocyte protein PMEL—which is expressed on melanoma cells. Its mechanism is to stimulate a cytotoxic T lymphocyte (CTL) response against tumor cells displaying the gp100 antigen. In clinical trials, gp100280-288 peptide is usually administered with an adjuvant (such as Montanide ISA-51 or QS-21), and sometimes in combination with a modified T-helper epitope from tetanus toxoid (AQYIKANSKFIGITEL) to enhance immunogenicity. The vaccine primarily targets high-risk resected or metastatic melanoma, with limited toxicity observed in studies. Immunological endpoints include induction of gp100-specific CD8+ T cell responses and helper T cell responses dominated by a Th1 cytokine profile.
