Overview

gp100:44-59 is a synthetic peptide epitope derived from the melanoma-associated antigen gp100, also known as Melanocyte protein PMEL or Pmel17. This specific 16-amino acid sequence (WNRQLYPEWTEAQRLD) corresponds to residues 44 through 59 of the full-length protein and serves as a major histocompatibility complex (MHC) Class II-restricted epitope. It is primarily recognized by CD4+ helper T cells when presented by HLA-DRB1*0401 molecules. In the context of cancer immunotherapy, gp100:44-59 is utilized as a therapeutic vaccine component to stimulate a helper T-cell response. By activating these gp100-specific CD4+ T cells, the peptide helps to orchestrate a more robust and durable anti-tumor immune environment, providing the necessary cytokine support (such as IFN-gamma and IL-2) to enhance the recruitment and effector function of CD8+ cytotoxic T cells against melanoma cells. Clinical development has historically been led by the National Cancer Institute (NCI) for the treatment of metastatic melanoma.
