Overview

GP11046 is a non-immunosuppressive immunophilin ligand originally developed by Guilford Pharmaceuticals. It is a small molecule analog of tacrolimus (FK506) designed to retain the neurotrophic and neuroregenerative properties of the parent compound without causing systemic immunosuppression. Unlike FK506, the GP11046-FKBP12 complex does not bind to or inhibit calcineurin, thereby avoiding the inhibition of T-cell activation. In the context of dermatology and hair loss, GP11046 has been investigated for its ability to stimulate hair follicle growth and modulate the hair cycle. Research led by Maria K. Hordinsky at the University of Minnesota has specifically evaluated its effects on human hair follicles grafted onto SCID mice to determine if its neurotrophic signaling can promote hair regrowth in conditions like alopecia areata.
